I want to add one director in Pvt Ltd company since 01/04/2012 and retire other one director from 05/11/2012. I want to know that can I file both the addition of director and retirement of director through One form 32.
or I have to file seperate forms..???

No, For filing Form-32 of details of more than one directors the date of event should be same.

In your case date of event is different in case of appointment and retirement, so you will file two Form-32.

10 January 2013 Two form 32 has to be filed since the gap between the event dates exceeds 30 days.
