why is revenu stamp of Rs 1 is affixed on payment voucher if payment exceeds Rs.5000 ? what it signifies ?

It is a requirement of the Stamp Act.

But what it signifies ? does it create any obligation or accountability on parties ?

The objective of the requirement is that the Government may earn reveune. That is all.
