Is there any rule prohibiting appointment of relatives as directors in an unlisted public company ( Being All directors appointed from one family only)?

Yes, in case of public company some provision requires Independent directors like constitution of Audit Committee, Remuneration committee, if such provision apply.
In that case, you can not appoint family members as Independent director to comply the above provisions.
