Kindly tell me sir whether the Proprietor of a business is liable to deduct tax at source while making payment of commission, which is in excess of Rs.2500

Individual should deduct TDS as per prescribed rates only if he is under tax audit.

agreed, if he is liable to tax audit , then he has to deduct tds .
