Does DTP applies for purchase from HUF concern (Consignment Agent of A Ltd) by proprietorship concern of kartha? HUF concern gets commission from A Ltd & it sells to many buyers including proprietorship concern of kartha. Purchases from HUF concern is 4.75 Crore before sales tax & including tax more than 5 Crore, does 5 Cr limit includes taxes also?

normally in income tax, revenue include taxes, so yes taxes should be included.
