A private company appointed relative of a director as the chief consultant, with the monthly remuneration exceeding Rs.250000/-, whether the Director Relative(place of Profit) Rules,2011 are applicale for the above appointment?

Yes,if such relative covered under Schedule IA of the Companies Act, 1956 then it will cover under section 314 of the Companies Act, 1956.
