Is it compulsary to take registration on intersate sale of excempted Goods?

Yes registration is required for inter state supply for exempt goods.

then what treatment of Freight paid to Unregistered Truck Owner and GTA . Please tell me in both case

GST not applicable on unregistered truck owners. For GTA you have to pay GST under RCM.
