IF A GOVT. EMPLOYEE PAID HOUSE RENT 3000/ PER MONTH, IS HE SUBMIT RENT RECEIPT TO D.D.O. OR NOT NECESSARY.

Yes he has to submit Rent receipt and other investment declaration based on which his TDS will be calculated and deducted.
