demand liabilty of income tax department regarding under sec 143 of computation of tax liability given by aseessee . what should be done and how to make reply to solve this case in income tax dept

if the computation as per 143 is wrong, you can file rectification request under section 154. this can be done online using your https://incometaxindiaefiling.gov.in/ login account.
