Resp. Experts I am salaried employee & got form 16 from my employer. Can I get deduction of Rs. 12500/- for medical expense incurred on treatment of my wife if actually the medical reimbursement is not given by employer.

Since you are a salaried employee and have Form 16, you can file ITR-1 (Sahaj) if your income is only from salary, one house property (without loss carried forward), and other sources like interest.
About medical expenses for your wife (Rs. 12,500) not reimbursed by employer — you cannot claim a deduction under medical reimbursement if your employer didn’t reimburse it.
However, if you paid for medical treatment under certain sections like 80D (health insurance premium) or 80DD/80U (for disabled dependents), you can claim deductions there, but normal medical expenses paid out-of-pocket are not deductible under Income Tax.
So, medical expenses paid for your wife without reimbursement are not allowed as a deduction in your ITR.
Summary:
File ITR-1 for your salary income.
Medical expenses paid personally (not reimbursed) — not deductible.
