Cost of acquisition of shares will include brokerage, stamp duty etc. Demat charges - i assume you are asking about demat account maintenance charges is disallowed as expense

In my trading account if i sell delivery shares from demat account a fixed charge of Rs 25 per company in a day is applicable and also brokerage and stamp duty is levied

IN MY TRADING LEDGER ACCOUNT IT SAYS "DP CHARGES FOR NSE /(BSE) SETTLEMENT NO 12345"

For calculation of short term gain for settlement No. 12345 the DP charges shall be deductible expense
