If assessee have only income from sale of Rural Agricultural Land. Whether it is mandatory to file income tax return?

Yes. File itr if u have agrrculture incime more than 2.5 lakh .Which need to be decleard in appropriate head of Itr.

No need to file ITR in case of such exempt income.
