If you are a GTA GST payable under RCM GST registration not required. If you are transporter of goods other than GTA such transport is exempt from GST, so GST registration not required.

As such GST registration is not mandatory if you are under RCM. But you can take registration if you want to claim ITC and charge gst on forward charge.
