i want to know that how capital gain is calculated on speculation income (same day-buy/sale of shares) Is Stt is taken in purchase cost & sale price...?

If STT is paid on securities then it will go to Exempted Income and will not b taxable as Capital Gain.

STT not include in sales price and purchase price.
