i have filled return original as well as revised now for original return 143(1) has been issued and rs.40000/- is increased and tax of rs. 4000/- is demanded?? should i file rectification or should wait for 143(1) of revised return????

In case you filed original return as belated return(i.e after the due date) then revised return doesn't consider and it is invalid. So in such case don't wait for 143(1) intimation for revised return because it never received by you because of CPC doesn't consider it so file rectification return.
In case you filed original return with in due date and filed revised return with in the process completion by the CPC then CPC only process your revised return and doesn't process your original return.
In case eventhough you filed original return with in due date but the CPC already processed your original return of you before to the revised return filed by you then it will again process your revised return.
So for the first or second case file rectification request immediately. In case of third case please wait for the 143(1) for the revised return.
I think your case may be first or third because of CPC processed your original return.
