We never charged depreciation on our fixed assets but want to charge it now ... Retrospective effect se karna hoga ?

Since you never provided depreciation on fixed assets but now you want to provide the same, you have to calculate the opening wdv of the fixed asset assuming if depreciation was charged from the beginning and the depreciation calculated from beginning to opening wdv would not be allowed as deduction
