According to the section 139(4C) for the education Institutions if their Total income exceeds basic exemption limit before giving effect u/s 10 provisions is required to file Income tax return.
So in your case if with out claiming exemption u/s 10 your total income exceeds basic exemption limit then required file Income tax return.
