Can anyone guide me whether is it mandatory that the proposed Directors in Form 1A should become Director in filing Form 32 for new company? There is a difference among proposed directors and so one director does not want to beome director and other person need to become director.

Yes, as and when you file Form-32, regarding appointment of director it becomes directo of that proposed company.
However, the name mentioned in Form-1A as proposed director can with draw there name to become director at the time of registration of the company and can vacate the post of deemed director.
