what is rule of 50 km Distance ???

No distance is irrelevant. you have to see consignment value and whether inter-state supply or intra-state supply. In some states ewaybill is to be generated for intra-state supplies too provided consignment value exceeds 50000/-.

Rule of 50km in regard to Eway bill , is no need to generate Part B , if the goods being send to transporter place for futher delivery (within state)
If intra-state ewaybill is required in your state then you have to generate ewaybill even if distance is less than 50km. Place of supply to consignee is to be given in ewaybill and not transporter. Part b is to be updated for change in conveyance
