My query is that is there difference between submission and filling of GSTR 3B.. As I submitted my GSTR 3B on 24th Aug. 2017 but due to DSC I can't filed it on 25th. am I supposed to pay penalty ??

You could have filed with EVC. OTP would have come to your registered Mobile and Email before filing. You can ask Helpcenter as to whether submission date is considered or filing date is considered for Penalty payment.
The OTP not applicable for firm or private limited company
