Is composition scheme available if there is purchase of goods inter state below 50000

Yes, it is available.
In composite scheme you can make inter state purchases without any limit. However inter-state sales are not allowed.
Secondly, limit of 50 lakhs has been revised up to 75 lakhs (except for 9 states)
But sir, as per section 10, stock held on appointed day should not include purchase from inter state.
