claiming of deduction
If we have not supplied information regarding the deduction to employer and so it is not shown in the form 16,then can we claim it in income tax return and get tds refund or is it compulsory that it should be in form 16?

Not Compulsory.
But you will be served a query by CPC u/s. 143(1)(A), for mismatch in the exemption claimed in ITR and form 16 or Form 26AS.
You will be required to submit proofs of the deduction claimed, and justify your claim within specified time.
