Actually we are deducting tds on prof for 2 parties for Rs.5000 & 4499 respectively. And dd taken for Rs.9449. We are filled the challan 281 for total Rs.9449. Can we give form 16 to 2 parties?

I think ur talking about non-salary deductees and in that case you have to issue Form 16A. And yes you can issue two separate Form 16A to two parties indicating the amount paid, amount deducted and all the challan details.
For format of Form 16A please visit www.incometaxindia.gov.in .
when you file TDs return and provide detail of TDS by Pan No. for both party in return. then you can issue form 16A to both party.

If in the TDS return (26Q) u have mentioned PAN no. of both the parties then only u can issue seprate Form 16A
