If a person has basic pay 25000, does he have the option to not get his PF deducted in an organisation on which PF act is applicable?

No.

Any employee who is already covered under EPF Act and due to Increment/Promotion if the wages is increased above than the ceiling limit (Rs. 15000/-) then the employee shall not be exempted. In this case both the contributions is restricted to Rs. 15000/- only.

 Any employee already had EPF account in his/her previous company and due to various reasons if the employee joins the new company and if the new company also has the EPF then that employee is covered under EPF act.

If the employee has joined an organisation for the 1st time in his life then does he have the option of not getting PF deducted?

No, if above ceiling limit....
