Yes rent agreement is mandatory not for claiming hra exemption.

Atleast rent receipt is required. Else it is better to make rent agreement

If you are a salaried employee then you must submit a receipt to your company.

If it's not there in form 16 and claiming additionally at the time of filing ITR then it is risky area and you must have legal document.
