if both the supplier and recipient are unregistered then is it mandatory to generate e-way bill.?

Yes, U will be generated when there is a movement of goods of value more than Rs. 50,000/ –

Not required as per rule only registred person is required to generate bill...

you has to enter ‘URP’ [Unregistered Person] in corresponding GSTIN column.
