does compulsory to prepare e waybill for inter state supply of amount less than rs.50000/- (for example rs.46000/-)?

NO.

ABOVE RS.50000 IT IS COMPULSORY FOR INTERSTATE SUPPLY

sorry there was misunderstanding.
my quary for transaction is not for export. so i reply it is not for export by mistake

in export there is require for way bill.

E-waybill is Mandatory for interstate trade irrespective of invoice value.
