For registration under MVAT act Voluntary the DD of Rs.25000/- and Rs.5000/- both has to be made or only make DD for Rs.25000/- can be sufficient and other Rs.5000/- can be paid by cash?

No it will be of 2 DD and not 1...
Such payment is to be made in by way of handing over the Demand Draft or Pay Order alongwith chalan in Form 210 to the registration officer. Demand draft or pay order is required to be prepared in favour of “Bank of Maharashtra – A/c. MVAT” in case of Mumbai region and “State Bank of India – A/c. MVAT” for other regions.
