If 2 companies have common directors and shareholders , but no company have any investment in capital of each other then is there is any relation of holding and subsidiary between them????

It depends...more than 50% of capital/voting rights holding is just one of the bases of establishing the holding -subsidiary relationship..
refer sec 2 (87) of the companies act 2013 for others.....
