If I am a proprietor of two separate business , do I require to get two gst no.

yes u will have to get separate registration.

Are they in the same state and is the nature of business same.

No Sir.,
If same state you ll be get One registration only...
Two or more states you can registered in all state and get different GSTN

Also if the party runs two business in different trade names having different trade license
