Is E way Bill mandatory if consignment value >50,000
but distance is only 2 km.

Is there any minimum distance requirement?

Every state has differnt rules it depends on state law in some states if it is within 2km than Part A is mandatory and can generate eway bill without part B in some states its no required.

No.... As per Rule 138 of CGST Act if consignment value exceeds 50k & being transported via motorised conveyance the Eway is Mandatory .

No need to Generate Part B only in case where the goods is transported to Transporter Place upto 50 km for further Delivery. (Notification 12/2017 CT)
