Deduction under section 80CCD (1B)
is there any requirement of contributing to tier 2 account for claiming exemption under section 80 CCD(1B)?

Whether you contribute to Tier-1 or 2 you can claim the deduction subject to maximum limit. There is no mandatory guidelines for the same.
