can we make changes in autopopulated gstr3b for monthly return fillers if there is diff in previous mth gstr3b sale.

Yes you can. GSTN will prompt you that the auto-populated value and your value is different, if you think you are correct and you have valid proofs to justify yourself then you can ignore the message sent by GSTN
